MOTS-c (Mitochondrial Open Reading Frame of the 12S rRNA type-c) is a synthetic 16-amino acid peptide derived from the mitochondrial genome and represents one of the first mitochondrial-derived peptides to be characterised in detail. Unlike most signalling peptides encoded by nuclear DNA, MOTS-c is encoded within the 12S ribosomal RNA region of mitochondrial DNA (mtDNA), highlighting the role of mitochondria as regulators of cellular communication in addition to energy production.

The discovery of MOTS-c significantly expanded scientific understanding of mitochondrial biology. Researchers continue investigating how mitochondrial-derived peptides regulate communication between mitochondria and the nucleus, influence cellular stress responses and contribute to metabolic homeostasis.

MOTS-c is synthesised from a short open reading frame located within mitochondrial 12S rRNA.
